Abstract: | Výsledkem této práce je další příspěvek ke studiu bakteriofágů. Poprvé byly bakteriofágy aplikovány do tekutého média, které vyžadují laboratorní kultury borrelií. Vytvořená a prakticky odzkoušená metodika se ukázala jako zcela použitelná, i když se fága lyzujícího borrelie zatím najít nepodařilo. Samotné studium problematiky bakteriofágů je důležité z vědeckého i praktického hlediska. Nabízí potenciál pro tvorbu efektivnějších léků v boji s mnoha bakteriálními infekcemi, na které ve vyšších stádiích nezabírají samotná antibiotika, protože se u nich vyvinula rezistence. Na tuto práci by proto měla navazovat další s větším záběrem možných hostitelů spirochét, kde se dají očekávat jejich viroví parazité. I získání mírně lyzujících fágů by bylo úspěchem, protože je možné jejich lyzační schopnosti různými technikami zvyšovat. V současné době přináší výzkum i dílčí výsledky, jako například rozpoznání parazitů střevních bakterií. |
Abstract in different language: | This thesis results in another contribution to the study of bacteriophages. A methodology, which has proven to be fully applicable, has been created and practically tested. For the first time, bacteriophages were applied to liquid media, which is required by borrelia laboratory cultures, and the bacteriophages were collected and examined. Although the main goal hasn't been achieved and borrelia lysing phages haven't been identified yet, the proof of the existence of bacteriophage DNA in cultures together with B. burgdorferi, and the absence of bacteriophages in pure B culture (see Figure 8), prove that this methodology is functional. The method of obtaining phages is appropriate, and it is clear that phages survive in B. burgdorferi cultures, even in liquid media in which B. burgdorferi is grown, although the phages used in the experiment don't directly attack B. burgdorferi. The study of bacteriophages is important from both scientific and practical point of view. It offers the potential to create more effective medicines to combat many bacterial infections that aren't cured by antibiotics alone in their higher stages because they have developed some resistance. |
